How It Works
Modafinil's exact mechanism is not fully understood, but it is believed to work primarily by inhibiting the reuptake of dopamine in the brain — increasing dopamine availability in synapses. It also influences norepinephrine, serotonin, histamine, and orexin (hypocretin) systems that regulate wakefulness and sleep-wake cycles. Unlike amphetamines, Modafinil does not cause widespread dopamine release, which explains its lower addiction potential and milder side effect profile. It promotes wakefulness by activating the hypothalamic wake-promoting regions while not suppressing REM sleep in the same way as traditional stimulants.
